Guarding the guardian: rescuing p53 mutants in cancer
The transcription factor p53 is the most frequently mutated protein in human cancer. Single amino acid substitutions in this tumor suppressor drive half of new cancer cases annually. One of the longstanding goals in cancer therapy is to develop drugs that restore transcriptional function to mutated p53. The Loh lab seeks to understand how common tumorigenic mutations cause p53 to lose function, and to develop drugs that interact with p53 to correct those defects. We focus on the role that zinc plays in p53 folding and misfolding, and how metal binding can be manipulated by small molecules to rescue mutant p53’s tumor killing abilities
